    Pi2, Pi9, Pi50, Pigm, Pizh, Piz, Piz-t 同為Piz位點上的復等位基因。其中,Fukunishiki 攜帶Pi2/Piz5,二八占攜帶Pi50,而Pi9來自小粒野生稻。

    【定位與克隆】

    Amante-Bordeos等首先將源自小粒野生稻中的抗稻瘟病基因導入栽培稻中,并命名為Pi9Pi9定位于水稻第6染色體,與之緊密連鎖的RFLP標記有RG64和R2132。Pi2初步定位在水稻第6 染色體上RG64 和RZ612 之間,遺傳距離分別是2.1 cM 和7.2 cM(Yu et al, 1991; Mew et al, 1994),后被精細定位在分子標記RG64 和AP22 之間,遺傳距離分別為0.9 cM 和1.2 cM(吳金紅等, 2002)。

    【抗譜表現】

    Pi9 抗譜相當廣,對來自13 個國家的43 個稻瘟病菌株均表現出很高的抗性(Liu et al, 2002);Pi2 抗譜也相當廣,對從中國收集的792 個稻瘟小種中的絕大部分表現抗性,只有7.55% 的小種能侵染攜帶Pi2 的親本C101A51(Chen et al, 2001),此外,C101A51對來自13 個國家的43 個稻瘟病菌株中的36 個表現抗性(Liu et al, 2002)。

    【基因的克隆與生物學功能分析】

    Pi9 區域含有6 個串聯排列的NBS-LRR 基因,其中Nbs2-Pi9Pi9Pi9 屬于NBS-LRR 類基因,含有2 個內含子,長度分別是5362bp 和128bp,Pi-9 cDNA 全長4009 bp,包含3099bp 的編碼區和910-bp 3' UTR。Pi-9 編碼一個由1032 氨基酸組成的蛋白產物,該產物羧基端含有富亮氨酸重復域(LRRs),由17 個不完整的LRR 重復組成;在氨基端含有NBS 域,NBS 域內有激酶1a、2和激酶3a等結構(Qu et al., 2006)。

    Pi9 在抗病植株中組成性表達,表達不受稻瘟病侵染誘導。Pi9 至少對21 個稻瘟病菌小種具有抗性,是一個稻瘟病廣譜抗性基因。Pi9 對稻瘟病的廣譜抗病性的分子機制可能是Pi9 可以識別不同稻瘟病菌小種的保守分子,或者Pi9 可以識別不同稻瘟病菌小種的不同分子。

    Pi2 屬于NBS-LRR類基因,含有2個內含子,長度分別是3839bp 和116bp,Pi2 cDNA 全長3332 bp,包含117-bp 5' UTR 和116-bp 3' UTR。Pi2 編碼產物包含1032 個氨基酸,含有1 個核苷酸結合位點(NBS)和3 個富亮氨酸重復區(LRR);Pi2Pi-zt 的編碼產物僅在3個LRRs區域有8個氨基酸的差異;而在這8突變氨基酸中,僅有1個位于xxLxLxx基序中(Zhou et al., 2006)。
